2.8 Koehler illumination

Koehler illumination needs to be adjusted in the beginning of every imaging session for optimal illumination. Proceed as follows:

1.Use 10x objective and front lens. Focus on the sample.

2.Close the luminous-field diaphragm by turning the adjustment wheel clockwise.

3.Move the condenser up or down so that the diaphragm edges are in focus. The microscope is designed so that in the imaging path, field diaphragm should be in conjugate plane with the specimen plane (in focus at the same time). You adjust this by adjusting the height of the condenser.
