Problem

Cause/Remedy

 

 

Phase Contrast

No phase contrast is possible.

￿The specimen is too thick, too thin or too brightly stained

￿Refractive index of mounting medium and specimen is identical so that there is no phase jump

￿The cover glass is not placed evenly ￿Check the right light ring (p. 44) ￿Check the centering of the light rings

(p. 29-30)

￿Check the condenser centering

￿Open the aperture diaphragm completely

Fluorescence

The image is completely dark (no fluorescence).

￿Open the shutter (p. 46)

 

￿Check the antigen-antibody combination

 

￿Insert a new lamp (p. 21 onwards)

The fluorescence is too weak.

Stage

￿Center the lamp (p. 31 onwards) ￿Insert a new lamp (p. 21 onwards)

Travel range of the stage in e-w direction decreases after a longer period of working.

￿Move the stage all the way to the left side with the coaxial pinion

￿Push one of the screws which hold the object guide with your hand further to the left as far as it will go

￿Afterwards move the stage all the way to the right

￿Push one of the screws which hold the object guide with your hand further to the right as far as it will go
